Comments |
The insert contains recognition sites for the following enzymes: HgiA1 (2), BglI (2), NarI, AvaI, PvuII (2), HindIII, EcoRI, PstI, StuI (2). A PstI digest will release a 1.1 kb fragment which contains the TNF coding sequence plus some vector sequence at the 5' end and some 3' untranslated sequence. Restriction digests of the clone give the following sizes (kb): PstI--2.4, 1.4, 1.1; HindIII--3.4, 1.5; BamHI--3.1, 1.8; EcoRI--4.6; EcoRI/PstI--2.4, 1.5, 1.0; EcoRI/BamHI--3.1, 1.1, 0.7; HindIII/PstI--2.4, 0.9, 0.8, 0.5; BamHI/HindIII--2.7, 1.0, 0.74, 0.47; StuI--3.5, 2.2, 0.25. |
