Restriction digests of the clone give the following sizes (kb): BamHI/HindIII--9.4, 1.5, 1.3; EcoRI/BamHI--6.3, 5.9; PstI--10.2, 2.0. IMPORTANT: To prevent amplification of a rearranged and/or deleted cosmid, we recommend streaking on LB + amp plates at 30C and picking small colonies for liquid culture. This is an integrative, positive selection vector conferring arginine prototrophy on an auxotrophic host such as Aspergillus niger 350(-)52 (ATCC 20739) (argB-). The argB BamHI/EcoRI fragment contains the following sites (separated by approximate kb): BamHI- 0.8- SalI- 0.4- HindIII- 1.4- SalI- 0.1- HindIII- 0.7- SalI- 0.1- HindIII- 0.7- SalI- 0.7- SalI- 0.5- EcoRI. This was constructed by inserting the 5.0 kb EcoRI/HindIII fragment from pBB116 containing argB into pHC79 cut with EcoRI + HindIII. |
