The insert contains the following restriction sites (separated by approximate kb): HindIII- 1.0- BamHI- 0.3- HindIII- 3.2- HindIII- 1.9- EcoRI- 0.25- BamHI- 1.1- EcoRI.
Restriction digests of the clone give the following sizes (kb): SmaI/SalI--8.6, 7.5, 2.7; SalI--16.0, 2.9; EcoRV--9.0, 6.2, 2.4, 1.0; XbaI--10.5, 4.1 (doublet).
A 1.3 kb HindIII fragment from this insert is sufficient to complement either a hem2-15 (C41/U1) or a hem2-16 (N28/U2) mutation.
The probe, pG32/T4, has been deposited in Escherichia coli RR1 as ATCC 37659 and in Saccharomyces cerevisiae C41/U1 as ATCC 66314.
