Restriction digests of the clone give the following sizes (kb): BamHI--6.6, 4.3; SacI--7.0, 3.0, 0.9. Shows transforming activity; insert contains 3 SacI sites (major fragments 3.0, 2.7, 0.85 kb), as well as XhoI, PvuII, XbaI, KpnI, SphI, ClaI, BglII sites; plasmid cut with SacI gives 7.0, 3.0, 0.85 kb fragments. This probe contains the transforming form of the HRAS1 gene mutated by a G to T transversion at the 12th codon; it also contains 3' and 5' flanking sequences. |
