The insert contains the following restriction sites, in bp from the 5' end of the insert: PstI (167), PstI (234), RsaI (285), XmnI (590). Restriction digests of the clone give the following sizes (kb): EcoRI--3.3, 0.9; BglI--1.5, 1.1 (doublet), 0.3; SphI--2.9, 1.1; BamHI--4.3; PvuI--4.3. The insert includes 49 bp of 5' untranslated, 480 bp encoding transforming growth factor alpha precursor, and 391 bp 3' untranslated. The RsaI polymorphism is in linkage disequilibrium with a BamHI polymorphism detected by phTGF1-10-3350, (r=0.55, p=0.03). Enzyme(s) not detecting polymorphism: BclII, BglII, EcoRI, EcoRV, HaeIII, HindIII, HinfI, KpnI, MspI, PstI, PvuII, SacI, XbaI, XhoI. |
