Protocol: ATCCNO: 50561 SPEC: This strain is routinely shipped as a frozen stabilate. Thaw the ampule and aseptically transfer the material to 10 ml of bacterized ATCC medium 1525 in a T-25 tissue culture flask. DO NOT distribute the thawed material to a larger volume of medium. It is essential to first establish the culture in a small volume. Screw cap on tightly and incubate horizontally at 25C. The culture should be ready to subculture in 5-7 days. To subculture, vigorously agitate the flask and aseptically transfer a 0.1 ml aliquot to 10 ml of bacterized ATCC medium 1525 containing a single rice grain and incubate as above. ATCC medium 1525 is bacterized by inoculating the medium with a loopful of Klebsiella pneumoniae subsp. pneumoniae ATCC 27889 (a non-pathogenic strain) from an ATCC medium 3 slant approximately 24 hours before using. Other strains of Klebsiella or other species of bacteria may serve equally well as a food source. This must be determined empirically.
