The construct contains unique StuI, EcoRI, and NdeI sites. Restriction digests of the clone give the following sizes (kb): EcoRI--4.0; BamHI/BglII--3.2, 0.88; NdeI--4.0. When expressed in Escherichia coli B121, this plasmid produces a protein that reacts with antibodies to transforming growth factor alpha and to Pseudomonas exotoxin A. This fusion protein has a molecular weight of 51 kDa. This was constructed by deleting some non-coding sequences from JH8 (ATCC 67208), adding an oligonucleotide containing a Tth111I site, and inserting a 152 bp Fnu4HI fragment from phTGF10-925 encoding TNFA into the Tth111I site. |
