Restriction digests of the clone give the following sizes (kb): EcoRI--6.6, 3.5; HindIII--6.0, 1.9, 1.4, 0.8; PstI--6.3, 3.8; KpnI--10.1. The insert contains the following restriction sites (approximate kb from the end upstream of ccrA): PstI--0.5, MboI--0.9, 1.9; KpnI--1.2; HindIII--3.6, 5.4. The 941 bp MboI fragment contains the complete gene, with 150 bp 5' and 50 bp 3' flanking sequence. The sequence of most of this fragment has been reported. The vector is a derivative of pMK16 deleted for the carboxy terminal half of the tetR marker and has had the small EcoRI/BamHI fragment replaced with the MCS-containing EcoRI/BamHI fragment from plink 322. |
