Restriction digests of the clone give the following sizes (kb): BamHI--3.7, 2.4; BglI--4.9, 1.2; SmaI--3.7, 2.4. pHE6 is a high-expression plasmid derived from pUC8. Inserts are expressed as run-on products initiated at lambda N. Synthesis is regulated by the cI857 repressor encoded by the plasmid. Corresponds to nt 109241-111960 of the GenBank record and includes a portion of the EBNA coding region (109298-109872, the carboxy-terminal third. Expresses a 28 kDa fusion protein composed of 36 amino acids from lambda N protein and linker and 191 amino acids of EBNA sequence. The protein is recognized by anti-EBNA antibodies and includes the DNA-binding domain that interacts with oriP. This SmaI fragment is downstream of the triplet repeat region in the BamHI K fragment. |
